What is Chenodeoxycholic Acid

Chenodeoxycholic acid is extracted from the bile of chicken, duck and goose. It is white or light yellow crystalline powder; it tastes bitter and has an unpleasant odor. Mp141-142°C. Easily soluble in ethanol, chloroform and glacial acetic acid, almost insoluble in water. This product can reduce the synthesis and secretion of cholesterol, reduce the excretion of total cholesterol in bile, thereby improving the solubility of cholesterol in bile, promoting the dissociation of cholesterol in stones, and achieving the stone-dissolving effect.

The effect of Chenodeoxycholic Acid:

Pharmacokinetics of Chenodeoxycholic Acid

Clinical chenodeoxycholic acid (CDCA) is highly chemically pure unconjugated cholic acid containing different crystalline forms. It dissolves quickly in the intestines and is almost completely absorbed in the usual dose of 500 to 750 mg, whether taken on an empty stomach or with food. A small part is bound to plasma proteins, and the free form in peripheral blood remains low. Part of it is discharged into the intestinal cavity through the bile duct and is reabsorbed, forming enterohepatic circulation. The liver can efficiently uptake and clear it, with a first-pass effect of 62%. CDCA is combined with glycine or taurine in the liver and secreted in bile. The combined CDCA can be reabsorbed and is more easily cleared by the liver. The bound form in the intestine can be free again and enter the newly ingested bile acid pool.But most of it is 7α- After dehydroxylation and decomposition, it becomes Lithocholic acid (LCA). LCA is the main metabolite after oral administration of CDCA. In normal humans, about 1/5 is absorbed through the terminal ileum and colon, and the rest is formed into bile salts and excreted in the feces. LCA is mainly sulfated in the liver and kidneys, reducing liver toxicity. Sulfate of LCA is rarely absorbed in the intestine and is excreted with feces, so sulfation can prevent the accumulation of LCA in the enterohepatic circulation.
