Introduction to Andrographolide

Andrographolide is an organic compound with the molecular formula C20H30O5 and white square prismatic or flaky crystals. The main active ingredient of the natural plant Andrographis paniculata has the effects of dispelling heat, detoxifying, anti-inflammatory and analgesic. It has special effects on bacterial and viral upper respiratory tract infections and dysentery, and is known as a natural antibiotic drug. This product is a diterpene lactone compound that is difficult to dissolve in water and can usually only be administered orally.

Preparation method of andrographolide

Andrographis paniculata leaves are soaked in 95% ethanol, and the resulting ethanol-soaked liquid is decolorized with activated carbon. The concentrated liquid after the decolorized liquid is distilled to recover the ethanol is left to obtain coarse crystals. Add 15 times the amount of 95% ethanol to the coarse crystals and heat to dissolve them. The activated carbon is decolorized and served while hot. Filter and leave to recrystallize to obtain a light yellow recrystallized product, which is then washed and refined with distilled water, chloroform, and methanol to obtain the finished product of andrographolide. For crude drugs, the total yield is 1-1.5%.

Andrographolide Detection Standard

The physical and chemical properties of Andrographolide

White square prismatic or flaky crystals (ethanol or methanol), odorless and bitter in taste.

Solubility: It is soluble in boiling ethanol, slightly soluble in methanol or ethanol, very slightly soluble in chloroform, and almost insoluble in water or ether. Melting point is 230~231℃.

Clinical application of Andrographolide

Andrographolide is used for bacillary dysentery, leptospirosis, meningitis, pneumonia, enhancement of adrenal cortex function, and upper respiratory tract infections. In response to the clinical needs of viral infection emergencies, different hydrophilic groups are introduced into the lactone structure of this product to enhance its water solubility and improve efficacy. A variety of andrographolide injections have been made from this. Chuanhuning injection, Yanhuning injection, and Lianbizhi injection, which are widely used clinically, are representative drugs of this type and have been approved by the Ministry of Health and the State Administration of Traditional Chinese Medicine. Listed as one of the essential drugs in the emergency department.
